Abstract
In this study, a polymer matrix based on microfibers obtained by the electrospinning technique was
prepared. Microfibers are characterised by their composition of polylactic acid (PLA) and
hydroxypropyl methyl cellulose (HPMC) which form a hybrid blend (PLA/HPMC). These fibres were
loaded with piperine, an active molecule with antitumor properties, which is hydrophilic in nature.
The main objective of this study was to establish the solubility of the polymers and the compatibility
of the solvents for optimization of the electrospinning process to obtain the matrices.
The electrospinning technique is a method that allows the production of fibres with a diameter of
less than 1 micron, which can be used in various applications, including drug delivery systems. The
electrospinning process involves the use of an electric field to draw polymer solutions or melts
through a small nozzle, resulting in the formation of microfibers. The electrospinning process
requires the use of solvents to dissolve the polymers, and the solubility of the polymers and the
compatibility of the solvents are critical parameters that affect the performance of the
electrospinning process.
To determine the solubility of the polymers, different solvents were used to dissolve PLA and HPMC.
The results showed that PLA was soluble in chloroform, while HPMC was soluble in water. The
compatibility of the solvents was also studied by mixing chloroform and water to determine if both
polymers could be dissolved in the same solvent. The results showed that the two polymers were not
compatible in the same solvent.
Therefore, the electrospinning process was optimised by using chloroform as a solvent for PLA and
water for HPMC. The different matrices prepared were characterised by the morphology of their
constituent fibres. The fibres were observed under a microscope, and it was found that the fibres
were smooth and had a diameter of less than 1 micron.
Finally, the study of piperine release from the fibre matrices was carried out. The release of piperine
from the matrices was determined by measuring the amount of piperine remaining in the matrices
after a certain period of time. The results showed that the piperine was released from the matrices
in a controlled manner, indicating that the PLA/HPMC microfibers can be used as a drug delivery
system.
In conclusion, the dual character (hydrophobic/hydrophilic) of PLA/HPMC microfibers allows the
loading of any drug, with the assurance of its compatibility and uniform distribution in the fibres, and
therefore could be considered a general drug delivery system. This study provides a new approach to
the production of microfibers using the electrospinning technique, which can be used in various
applications, including drug delivery systems.
