Abstract
Diabetes is an ongoing huge global health issue, which is predicted to affect 552 million people worldwide in 2030. In fact, glucose monitoring biosensors account for 85% of the entire biosensor market. The demand for non-invasive glucose level monitoring is large and numerous research towards a simple, reliable and cheap biosensor is being carried out. This project is a continuation of various studies. Previous research showed the possibility to construct an enzymatic electrochemical biosensor with plasma-treated low-density polyethylene (LDPE). Moreover, glucose oxidase (GOx) was adsorbed onto the biosensor surface by means of a covalent bond in order to promote the detection of glucose. Plasma treatment proved to be a reliable method for the production of a vast variety of reactive species. The adsorption of these species on the electrode surface catalyse the glucose oxidation substantially improving its detection. Thus, a logical next step to take so as to improve the biosensor performance is to vary the plasma treating conditions. The goal of this project is to identify and characterise the most suiting plasma ambient for the biosensor surface modification. In a first part, biosensor based on the immobilization of GOx onto LDPE is produced. The utilisation of 1-(3-(dimethylamino) propyl)-3-ethylcarbodiimide methiodide as an activating agent for the coupling of amines is fundamental for the enzyme immobilization to thrive. Afterwards, electrochemical techniques are employed to characterise the biosensor efficacy. Next, 10 different plasma ambients are used on the electrode surface. Plasma is applied for 1min on each electrode. Later on, changes in the surface are detected and described using X-ray photoelectron spectroscopy (XPS). Finally, discussion over the efficacy of the different plasma ambients method is argued Investigaciones previas han demostrado la posibilidad de construir un biosensor electroquímico enzimático con LDPE tratado con plasma. El sensor trabaja mediante la glucosa oxidasa, que está adsorbida en la superficie del biosensor mediante un enlace covalente para promover la detección de glucosa. El objetivo de este proyecto es identificar y caracterizar diferentes ambientes de plasma con los cuales se ha tratado previamente la superficie del electrodo mediante XPS y métodos electroquímicos. Investigacions prèvies han demostrat la possibilitat de construir un biosensor electroquímic enzimàtic amb LDPE tractat amb plasma. El sensor treballa mitjançant la glucosa oxidasa, que està absorbida a la superfície del biosensor mitjançant un enllaç covalent per promoure la detecció de glucosa. L'objectiu d'aquest projecte és identificar i caracteritzar diferents ambients de plasma amb els quals s'ha tractat la superfície de l'electrode mitjançant XPS i mètodes electroquímics.
