Statistical efficiency of survival endpoints in breast cancer clinical trials

Abstract

Per demostrar l'eficàia d'una nova teràpia, en assajos clínics en càncer es recomana l'ús d'endpoints basats en la supervivència. D'entre ells, les principals agències reguladores (FDA i EMA) recomanen l'ús d' Overall Survival, entre d'altres raóns perquè és fàcilment observable. En aquest estudi, basats en la metodología descrita per Gomez i Lagakos, estudiarem l'eficiència estadística d'aquests endpoints en funció de la probabilitat dels events, el hazard ratio i la seva correlació. Analitzarem mitjançant l'eficiència relativa assimptòtica, en quines condicions l'ús de mort estarà recomanat, i en quines afegint progressió, i creant un composite endpoint (progressió o mort) tindrem un endpoint més eficient estadísticament parlant. . The objective of this Master Thesis will be, through the use of the asymptotic relative efficiency (ARE) , to evaluate in which cases the use of Overall Survival (OS) as main endpoint is more efficient to detect differences in a given breast cancer randomized trial, and in which cases the addition of progression will be beneficial.The student will have to: 1) learn the method of Gómez and Lagakos (2012); 2) perform a search of clinical trials on breast cancer and identify the values of the parameters needed to compute the ARE; and 3) set specific recommendations
